[Prediction of drug-induced hepatotoxicity using microsomal fraction homogenates].
The models of in vitro hepatotoxicity were developed in the study to predict the drug-induced hepatotoxicity. The model of in vivo hepatotoxicity was developed by determining the potential of the drug to induce hepatotoxicity using the models of in vitro hepatotoxicity in animals. Seventy-two commonly used drugs known to induce hepatotoxicity were selected as test drugs for the model of in vitro hepatotoxicity. The results showed that the models for the class III drugs had better prediction of hepatotoxicity than the models for the other classes of drugs, and that the model for the class III drugs had good predictive power of hepatotoxicity. The prediction for hepatotoxicity in the humans was evaluated by comparing these models with the in vivo hepatotoxicity data of the test drugs obtained from the database of the US-FDA. The results showed that the models predicted the hepatotoxicity in the humans well. Further, the models were applied to the prediction of toxicities in the humans. The prediction results showed that 14 in vitro models for the class III
