Ample research has shown that schizophrenia is characterized by abnormalities in the recognition of emotional expressions. We have recently shown that brain response to face expressions in the amygdala is attenuated in chronic schizophrenia. We have also shown that ketamine, an NMDA receptor antagonist, has a facilitating effect on the amygdala response in schizophrenia. The present study investigated whether ketamine could improve face emotion recognition deficits in schizophrenia. We used functional magnetic resonance imaging to evaluate the effects of ketamine on the brain response to emotional faces in 21 schizophrenia patients and in 21 healthy volunteers. Emotional expressions were presented before and after ketamine administration. The whole-brain contrast analyses revealed that the effect of ketamine was most pronounced in the left amygdala. The significant effect of ketamine was significantly associated with the social subscale of the PANSS. The whole-brain contrast analysis revealed that the combined effect of ketamine and emotional expression enhanced the brain response to emotional faces in the amygdala in healthy volunteers, which was not the case in schizophrenia patients.
