The safe and effective use of the paracetamol (Trade Mark) and ibuprofen (Trade Mark) prescribed medicines was evaluated through determining the gastric emptying, non-invasive permeability, and the oral absorption in healthy male subjects. The volunteer subjects were recruited from the graduate research volunteers at the Faculty of Pharmacy, Universitas Padjadjaran, Indonesia. Three capsules of 2.5 grams were taken, which consisted of 500 mg of paracetamol or 150 mg of ibuprofen, and 30 ml of aqueous solution of 9% hyaluronic acid or isotonic solution of 5% glucose in 10 ml of water. The pH of the solutions ranged from 2.8 to 3.5. The samples were prepared after 45 minutes of the volunteers fasting. The gastric emptying and the absorption phases were determined with the use of a breath hydrogen analysis using a method that was previously validated. The oral absorption phase was calculated with the use of the drug bioavailability programme designed in the basic research laboratory of the Faculty of Pharmacy. The motility phases of the volunteers were checked through the measuring of the gastric emptying time. The noninvasive permeability phase was performed according to Food and Drug Administration (FDA) of the USA through a method that was validated to measure the drug absorption in the absorption phases. All the tests were performed in the fasting state. The bioavailability of both the drugs in these capsule formulations were determined to be 45% after one hour of administration, and this value was used in the calculation of the permeability phase to predict the drug absorption. The bioavailability of paracetamol (Trade Mark) and ibuprofen (Trade Mark) were found to be 45% and 47%, respectively.
